Remove the swab from the saline and depress onto two (2) areas of a clean, dry microscope slide expressing a small amount of fluid on each. To one sample area of the slide, add a drop of KOH. 3% potassium hydroxide (KOH) solution (3 grams KOH per mL distilled water) on a clean glass slide. B. Stir for about one minute, occasionally lifting the loop to look for thickening and “stringing” of the slurry. C. RESULTS: 1. Gram positive bacteria will not appear to change the viscosity of the KOH solution. 2.

Procedure Specimen collection Skin. Cleanse skin to be sampled with 70% alcohol to remove contaminating microoranisms. If a characteristic dermatophyte “ring” is present on the skin, collect samples from the outer margin of the ring at its junction with the normal skin Otherwise collect samples from areas where the skin apperats to be scaling. Procedure 1. Place a portion of the specimen on the slide (select a purulent area if secretions). 2. Add 1 to 2 drops KOH and emulsify specimen. If it does not clear rapidly, place slide in petri dish and allow to stand about 10 minutes. 3. If tissue or scrapings, place the slide on C bench top heating block for minutes to speed clearing. 4. 3% potassium hydroxide (KOH) solution (3 grams KOH per mL distilled water) on a clean glass slide. B. Stir for about one minute, occasionally lifting the loop to look for thickening and “stringing” of the slurry. C. RESULTS: 1. Gram positive bacteria will not appear to change the viscosity of the KOH solution. 2.
